NM_004336.5(BUB1):c.469A>G (p.Arg157Gly) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R157G variant (also known as c.469A>G), located in coding exon 6 of the BUB1 gene, results from an A to G substitution at nucleotide position 469. The arginine at codon 157 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
