This variant lies in the SMARCA4 gene (transcript NM_003072.5) at coding-DNA position 4584, where C is replaced by A; at the protein level this means replaces aspartic acid at residue 1528 with glutamic acid — a missense variant. Submitter rationale: The p.D1560E variant (also known as c.4680C>A), located in coding exon 32 of the SMARCA4 gene, results from a C to A substitution at nucleotide position 4680. The aspartic acid at codon 1560 is replaced by glutamic acid, an amino acid with highly similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of p.D1560E remains unclear.
