Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.4729C>G (p.Gln1577Glu), citing Ambry Variant Classification Scheme 2023: The p.Q1556E variant (also known as c.4666C>G), located in coding exon 35 of the NF1 gene, results from a C to G substitution at nucleotide position 4666. The glutamine at codon 1556 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
