NM_001105206.3(LAMA4):c.4652G>A (p.Gly1551Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G1544D variant (also known as c.4631G>A), located in coding exon 32 of the LAMA4 gene, results from a G to A substitution at nucleotide position 4631. The glycine at codon 1544 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species; however, aspartic acid is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
