Uncertain significance — the classification assigned by Ambry Genetics to NM_024642.5(GALNT12):c.461G>T (p.Arg154Leu), citing Ambry Variant Classification Scheme 2023: The p.R154L variant (also known as c.461G>T), located in coding exon 2 of the GALNT12 gene, results from a G to T substitution at nucleotide position 461. The arginine at codon 154 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
