Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_015450.3(POT1):c.461G>A (p.Cys154Tyr), citing Ambry Variant Classification Scheme 2023: The p.C154Y variant (also known as c.461G>A), located in coding exon 4 of the POT1 gene, results from a G to A substitution at nucleotide position 461. The cysteine at codon 154 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
