Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.4681A>G (p.Ser1561Gly), citing Ambry Variant Classification Scheme 2023: The p.S1540G variant (also known as c.4618A>G), located in coding exon 34 of the NF1 gene, results from an A to G substitution at nucleotide position 4618. The serine at codon 1540 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
