NM_001365951.3(KIF1B):c.4748C>T (p.Thr1583Ile) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T1537I variant (also known as c.4610C>T), located in coding exon 40 of the KIF1B gene, results from a C to T substitution at nucleotide position 4610. The threonine at codon 1537 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
