NM_177438.3(DICER1):c.4600A>C (p.Asn1534His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N1534H variant (also known as c.4600A>C), located in coding exon 22 of the DICER1 gene, results from an A to C substitution at nucleotide position 4600. The asparagine at codon 1534 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
