Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001048174.2(MUTYH):c.3G>T (p.Met1Ile), citing Ambry Variant Classification Scheme 2023: The p.M15I variant (also known as c.45G>T), located in coding exon 2 of the MUTYH gene, results from a G to T substitution at nucleotide position 45. The methionine at codon 15 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
