Uncertain significance — the classification assigned by Ambry Genetics to NM_001387690.1(KATNAL2):c.1393G>C (p.Gly465Arg), citing Ambry Variant Classification Scheme 2023: The p.G393R variant (also known as c.1177G>C), located in coding exon 13 of the KATNAL2 gene, results from a G to C substitution at nucleotide position 1177. The glycine at codon 393 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
