Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004064.5(CDKN1B):c.449G>T (p.Gly150Val), citing Ambry Variant Classification Scheme 2023: The p.G150V variant (also known as c.449G>T), located in coding exon 1 of the CDKN1B gene, results from a G to T substitution at nucleotide position 449. The glycine at codon 150 is replaced by valine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
