Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003036.4(SKI):c.1177C>A (p.Leu393Ile), citing Ambry Variant Classification Scheme 2023: The p.L393I variant (also known as c.1177C>A), located in coding exon 3 of the SKI gene, results from a C to A substitution at nucleotide position 1177. The leucine at codon 393 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
