NM_003072.5(SMARCA4):c.4399G>A (p.Asp1467Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1499N variant (also known as c.4495G>A), located in coding exon 30 of the SMARCA4 gene, results from a G to A substitution at nucleotide position 4495. The aspartic acid at codon 1499 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
