Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_002875.5(RAD51):c.445G>T (p.Asp149Tyr), citing Ambry Variant Classification Scheme 2023: The p.D149Y variant (also known as c.445G>T), located in coding exon 5 of the RAD51 gene, results from a G to T substitution at nucleotide position 445. The aspartic acid at codon 149 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
