This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 445, where G is replaced by C; at the protein level this means replaces glycine at residue 149 with arginine — a missense variant. Submitter rationale: The p.G149R variant (also known as c.445G>C), located in coding exon 3 of the MSH2 gene, results from a G to C substitution at nucleotide position 445. The glycine at codon 149 is replaced by arginine, an amino acid with dissimilar properties. In a massively parallel cell-based functional assay testing susceptibility to a DNA damaging agent, 6-thioguanine (6-TG), this variant was reported to be functionally deleterious (Jia X et al. Am J Hum Genet, 2021 01;108:163-175). Based on internal structural analysis, p.G149R is highly destabilizing to the local structure (Ambry internal data).
