Likely pathogenic for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000138.5(FBN1):c.4459+1del, citing Ambry Variant Classification Scheme 2023: The c.4459+1delG intronic variant, located in intron 35 of the FBN1 gene, results from a deletion of one nucleotide one nucleotide after coding exon 35 of the FBN1 gene. This nucleotide position is highly conserved in available vertebrate species. Using the BDGP and ESEfinder splice site prediction tools, this alteration is predicted to weaken the efficiency of the native splice donor site; however, direct evidence is unavailable. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay. As such, this alteration is classified as likely pathogenic.
