NM_000138.5(FBN1):c.4423G>C (p.Gly1475Arg) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G1475R variant (also known as c.4423G>C), located in coding exon 35 of the FBN1 gene, results from a G to C substitution at nucleotide position 4423. The glycine at codon 1475 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
