Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.4412G>C (p.Arg1471Thr), citing Ambry Variant Classification Scheme 2023: The p.R1471T variant (also known as c.4412G>C), located in coding exon 33 of the TSC2 gene, results from a G to C substitution at nucleotide position 4412. The arginine at codon 1471 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
