Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.439C>G (p.Gln147Glu), citing Ambry Variant Classification Scheme 2023: The p.Q147E variant (also known as c.439C>G), located in coding exon 4 of the RECQL gene, results from a C to G substitution at nucleotide position 439. The glutamine at codon 147 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
