NM_016507.4(CDK12):c.4382dup (p.Thr1463fs) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.4382dupG variant, located in coding exon 14 of the CDK12 gene, results from a duplication of G at nucleotide position 4382, causing a translational frameshift with a predicted alternate stop codon (p.T1463Nfs*50). This frameshift occurs at the 3' terminus of the CDK12 gene, is not expected to trigger nonsense-mediated mRNA decay, and impacts only the last 28 amino acids of the protein. The exact functional impact of these altered amino acids is unknown at this time. Additionally, loss of function of CDK12 has not been clearly established as a mechanism of disease. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
