Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.4516G>C (p.Glu1506Gln), citing Ambry Variant Classification Scheme 2023: The p.E1460Q variant (also known as c.4378G>C), located in coding exon 40 of the KIF1B gene, results from a G to C substitution at nucleotide position 4378. The glutamic acid at codon 1460 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
