Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_003002.4(SDHD):c.435C>A (p.His145Gln), citing Ambry Variant Classification Scheme 2023: The p.H145Q variant (also known as c.435C>A), located in coding exon 4 of the SDHD gene, results from a C to A substitution at nucleotide position 435. The histidine at codon 145 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
