NM_001365951.3(KIF1B):c.4472A>T (p.Glu1491Val) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1445V variant (also known as c.4334A>T), located in coding exon 39 of the KIF1B gene, results from an A to T substitution at nucleotide position 4334. The glutamic acid at codon 1445 is replaced by valine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
