NM_000492.4(CFTR):c.4323C>G (p.Ile1441Met) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I1441M variant (also known as c.4323C>G), located in coding exon 27 of the CFTR gene, results from a C to G substitution at nucleotide position 4323. The isoleucine at codon 1441 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
