Uncertain significance — the classification assigned by Ambry Genetics to NM_024642.5(GALNT12):c.430T>C (p.Phe144Leu), citing Ambry Variant Classification Scheme 2023: The p.F144L variant (also known as c.430T>C), located in coding exon 2 of the GALNT12 gene, results from a T to C substitution at nucleotide position 430. The phenylalanine at codon 144 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
