Submitter rationale: The p.D1434N variant (also known as c.4300G>A), located in coding exon 2 of the ZNF469 gene, results from a G to A substitution at nucleotide position 4300. The aspartic acid at codon 1434 is replaced by asparagine, an amino acid with highly similar properties. This alteration was reported in a keratoconus cohort (Yu X et al. Mol Vis, 2017 Apr;23:296-305). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
