Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001291867.2(NHS):c.4350-5C>G, citing Ambry Variant Classification Scheme 2023: The c.4287-5C>G intronic variant results from a C to G substitution 5 nucleotides upstream from coding exon 8 in the NHS gene. This nucleotide position is poorly conserved in available vertebrate species. Using two different splice site prediction tools, this alteration is predicted by ESEfinder to abolish the native splice acceptor site, but is predicted to weaken (but not abolish) the efficiency of the native splice acceptor site by BDGP; however, direct evidence is unavailable. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
