Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.4285T>C (p.Tyr1429His), citing Ambry Variant Classification Scheme 2023: The p.Y1429H variant (also known as c.4285T>C), located in coding exon 11 of the BRCA1 gene, results from a T to C substitution at nucleotide position 4285. The tyrosine at codon 1429 is replaced by histidine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
