Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.4264GAG[2] (p.Glu1424_Glu1425del), citing Ambry Variant Classification Scheme 2023: The c.4270_4275delGAGGAG variant (also known as p.E1424_E1425del) is located in coding exon 22 of the DICER1 gene. This variant results from an in-frame GAGGAG deletion at nucleotide positions 4270 to 4275. This results in the in-frame deletion of two glutamic acid residues at codons 1424 to 1425. These amino acid positions are not well conserved in available vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
