NM_001999.4(FBN2):c.4267G>T (p.Ala1423Ser) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A1423S variant (also known as c.4267G>T), located in coding exon 33 of the FBN2 gene, results from a G to T substitution at nucleotide position 4267. The alanine at codon 1423 is replaced by serine, an amino acid with similar properties, and is located in a cbEGF-like domain. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
