Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.4396G>A (p.Asp1466Asn), citing Ambry Variant Classification Scheme 2023: The p.D1420N variant (also known as c.4258G>A), located in coding exon 39 of the KIF1B gene, results from a G to A substitution at nucleotide position 4258. The aspartic acid at codon 1420 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
