NM_004304.5(ALK):c.424G>C (p.Val142Leu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V142L variant (also known as c.424G>C), located in coding exon 1 of the ALK gene, results from a G to C substitution at nucleotide position 424. The valine at codon 142 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
