NM_024301.5(FKRP):c.424G>A (p.Glu142Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E142K variant (also known as c.424G>A), located in coding exon 1 of the FKRP gene, results from a G to A substitution at nucleotide position 424. The glutamic acid at codon 142 is replaced by lysine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species, and lysine is the reference amino acid in other vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
