NM_004304.5(ALK):c.4249G>T (p.Asp1417Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1417Y variant (also known as c.4249G>T), located in coding exon 29 of the ALK gene, results from a G to T substitution at nucleotide position 4249. The aspartic acid at codon 1417 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
