Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.4246T>C (p.Tyr1416His), citing Ambry Variant Classification Scheme 2023: The p.Y1416H variant (also known as c.4246T>C), located in coding exon 27 of the DNAH5 gene, results from a T to C substitution at nucleotide position 4246. The tyrosine at codon 1416 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
