NM_001048174.2(MUTYH):c.339G>C (p.Gln113His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q141H variant (also known as c.423G>C), located in coding exon 5 of the MUTYH gene, results from a G to C substitution at nucleotide position 423. The glutamine at codon 141 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
