NM_001267550.2(TTN):c.69541A>G (p.Lys23181Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K14116E variant (also known as c.42346A>G), located in coding exon 152 of the TTN gene, results from an A to G substitution at nucleotide position 42346. The lysine at codon 14116 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
