NM_001148.6(ANK2):c.11693A>T (p.Lys3898Met) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K3898M variant (also known as c.11693A>T), located in coding exon 44 of the ANK2 gene, results from an A to T substitution at nucleotide position 11693. The lysine at codon 3898 is replaced by methionine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
