Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.4358G>C (p.Gly1453Ala), citing Ambry Variant Classification Scheme 2023: The p.G1407A variant (also known as c.4220G>C), located in coding exon 38 of the KIF1B gene, results from a G to C substitution at nucleotide position 4220. The glycine at codon 1407 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
