NM_006361.6(HOXB13):c.421G>C (p.Ala141Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A141P variant (also known as c.421G>C), located in coding exon 1 of the HOXB13 gene, results from a G to C substitution at nucleotide position 421. The alanine at codon 141 is replaced by proline, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
