This variant lies in the RECQL gene (transcript NM_002907.4) at coding-DNA position 421, where A is replaced by T; at the protein level this means replaces isoleucine at residue 141 with phenylalanine — a missense variant. Submitter rationale: The p.I141F variant (also known as c.421A>T), located in coding exon 4 of the RECQL gene, results from an A to T substitution at nucleotide position 421. The isoleucine at codon 141 is replaced by phenylalanine, an amino acid with highly similar properties. This variant was identified in 1/302 breast cancer patients who tested negative for BRCA1/2 mutations and was not detected in 250 controls. The carrier of this alteration was diagnosed at age 47 and had family history of a mother diagnosed with breast cancer at age 70 (Rashid MU et al. Hered Cancer Clin Pract, 2020 Dec;18:25). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
