Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.421A>G (p.Arg141Gly), citing Ambry Variant Classification Scheme 2023: The p.R141G variant (also known as c.421A>G), located in coding exon 4 of the BUB1 gene, results from an A to G substitution at nucleotide position 421. The arginine at codon 141 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
