Uncertain significance for Hereditary cancer-predisposing syndrome; Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_005359.6(SMAD4):c.1000C>G (p.Gln334Glu), citing Ambry Variant Classification Scheme 2023: The p.Q334E variant (also known as c.1000C>G), located in coding exon 8 of the SMAD4 gene, results from a C to G substitution at nucleotide position 1000. The glutamine at codon 334 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
