Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001010892.3(RSPH4A):c.418C>A (p.His140Asn), citing Ambry Variant Classification Scheme 2023: The p.H140N variant (also known as c.418C>A), located in coding exon 1 of the RSPH4A gene, results from a C to A substitution at nucleotide position 418. The histidine at codon 140 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
