Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001148.6(ANK2):c.4180A>G (p.Lys1394Glu), citing Ambry Variant Classification Scheme 2023: The p.K1394E variant (also known as c.4180A>G), located in coding exon 34 of the ANK2 gene, results from an A to G substitution at nucleotide position 4180. The lysine at codon 1394 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
