Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.4180A>C (p.Ser1394Arg), citing Ambry Variant Classification Scheme 2023: The p.S1394R variant (also known as c.4180A>C), located in coding exon 32 of the FBN2 gene, results from an A to C substitution at nucleotide position 4180. The serine at codon 1394 is replaced by arginine, an amino acid with dissimilar properties, and is located in the cbEGF-like #19 domain. This amino acid position is not well conserved in available vertebrate species, and arginine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
