Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001267550.2(TTN):c.68819A>G (p.Glu22940Gly), citing Ambry Variant Classification Scheme 2023: The p.E13875G variant (also known as c.41624A>G), located in coding exon 150 of the TTN gene, results from an A to G substitution at nucleotide position 41624. The glutamic acid at codon 13875 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
