This variant lies in the CFTR gene (transcript NM_000492.4) at coding-DNA position 415, where C is replaced by T; at the protein level this means replaces histidine at residue 139 with tyrosine — a missense variant. Submitter rationale: The p.H139Y variant (also known as c.415C>T), located in coding exon 4 of the CFTR gene, results from a C to T substitution at nucleotide position 415. The histidine at codon 139 is replaced by tyrosine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is not well conserved in available vertebrate species.
