Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.4133G>C (p.Gly1378Ala), citing Ambry Variant Classification Scheme 2023: The p.G1378A variant (also known as c.4133G>C), located in coding exon 21 of the BLM gene, results from a G to C substitution at nucleotide position 4133. The glycine at codon 1378 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
